Use | 3, 5-dichloropyrazine-2-formic acid is an organic intermediate that can be used as an intermediate to prepare perfluorinated cyclopropyl fused 1, 3-oxazine-2-amine compounds of β secretase inhibitors. |
preparation | 2,6-dichloropyrazine (1.44g) was added to the solution of lithium diisopropylamide (2.0M heptane/tetrahydrofuran/ethylbenzene, 11.10mL,22.20mmol) in THF(75mL) at -78 ℃ for 20min at room temperature, 9.67mmol) in THF(20mL). The reaction mixture was stirred at -78°C for 1.5h and added via a sleeve to a three-neck flask containing dry ice at -78°C. The reaction mixture was heated from -78°C to room temperature over 21h and quenched with 5MHCl. Distribute the mixture between brine and EtOAc. The aqueous phase was acidified to pH 3.5 with 5M HCl. The aqueous phase was extracted with EtOAc(6x), and the combined organic extract was washed with brine (1x), dried with MgSO4, filtered and concentrated. 3, 5-dichloropyrazine-2-formic acid (0.408g,2.11mmol, 22% yield) in a light brown solid form was obtained by silica gel rapid column chromatography (5% to 10% MeOH in DCM). LC/MS(ESI )m/z = 193.0(M H). |
